Abstract
The aim of this study was to examine the staining characterstics of lipofuscin in heart, midgut and malpighian tubule of ageing male D. similis. Samples of different organs of 5, 10, 15, 20 and 25 days old insects were examined with various stains under light microscope. Paraffin sections cut at 6 p., thickness and were stained with Nile blue A, ferric ferricyanide and carbolfuchsin methods. Histochemical results in the heart, midgut and malpighian tubule of ageing male D. similis revealed that the pigment granules showed characteristically different staining behaviour with different stains at different age levels. It was clearly noted that one stain had strong affinities with the pigment granules of one tissue at a certain age levels, while the same stain had mild or moderate affinities with pigments of the same tissue at different age levels.
